A new genetic mutation that causes familial amyotrophic lateral sclerosis (ALS), a fatal neurological disorder also known as Lou Gehrig's Disease, has been identified by a team of scientists led by researchers at the University of Massachusetts Medical School (UMMS). Mutations to the profilin (PFN1) gene, which is essential to the growth and development of nerve cell axons, is estimated to account for one to two percent of inherited ALS cases. The finding, described July 15 in the online edition of Nature, points to defects in a neuron's cytoskeleton structure as a potential common feature among diverse ALS genes.
"This discovery identifies what may possibly be a common biological mechanism involved across familial ALS cases regardless of genetics," said John Landers, PhD, associate professor of neurology and senior author of the study. "We know of at least three other ALS genes, in addition to PFN1, that adversely impact axon growth. If indeed, this is part of the disease's mechanism, then it might also be a potential target for therapeutics."
Robert Brown, MD, DPhil, a co-author on the study and chair of neurology at UMass Medical School, said "Dr. Landers has done great work in defining this new pathway for motor neuron death. We are delighted to have identified the defects in families from the U.S., Israel and France that we have been investigating for several years. Our finding is particularly exciting because it may provide new insights into ALS treatment targets."
ALS is a progressive, neurodegenerative disorder affecting the motor neurons in the central nervous system. As motor neurons die, the brain's ability to send signals to the body's muscles is compromised. This leads to loss of voluntary muscle movement, paralysis and eventually respiratory failure. The cause of most cases of ALS is not known. Approximately 10 percent of cases are inherited. Though investigators at UMass Medical School and elsewhere have identified several genes shown to cause inherited or familial ALS, almost 50 percent of these cases have an unknown genetic cause.
The current Nature study details the discovery of the PFN1 gene mutation among two large ALS families. Both families were negative for known ALS-causing mutations and displayed familial relationships that suggested a dominant inheritance mode for the disease. For each family, two affected members with maximum genetic distance were selected for deep DNA sequencing. To identify an ALS-causing mutation, genetic variations between the family members were identified and screened against known databases of human genetic variation, such as the 1000 Genomes Project. This narrowed down the resulting number of candidate, ALS-causing mutations to two within the first family and three within the second. Interestingly, both families contained different mutations within the same gene -- PFN1, the likely causative mutation. With additional screening, the team documented that in a total of 274 families sequenced, seven contained a mutation to the PFN1 gene, establishing it as a likely cause for ALS.
While it is not certain how the PFN1 mutation causes ALS, the cellular functions it controls within the motor neurons are responsible for regulation of a number of activities, including the growth and development of the axon, the slender projection through which neurons transmit electrical impulses to neighboring cells, such as muscle. When introduced into motor neuron cells, normal PFN1 protein was found diffused throughout the cytoplasm. Conversely, the mutant PFN1 observed in ALS patients was found to collect in dense aggregates, keeping it from functioning properly. Motor neurons producing mutated PFN1 showed markedly shorter axon outgrowth.
"The discovery that mutant PFN1 interferes with axon outgrowth was very exciting to us," said Claudia Fallini, PhD, a postdoctoral researcher at Emory University School of Medicine who collaborated with the UMass Medical School authors to investigate PFN1's functions in cultured motor neurons. "It suggests that alterations in actin dynamics may be an important mechanism at the basis of motor neuron degeneration."
"In healthy neurons, PFN1 acts almost like a railroad tie for fibrous filaments called actin, which make up the axon" said Landers. "PFN1 helps bind these filaments to each other, promoting outgrowth of the axon. Without properly functioning PFN1 these filaments can't come together. Here we show that mutant PFN1 may contribute to ALS pathogeneses by accumulating in these aggregates and altering the actin dynamics in a way that inhibits axon outgrowth."

"Yahoo! Voices' administrators made a big mistake storing the passwords in plaintext, but all users need to bolster their own security as well. Make passwords harder to guess by making them more than eight characters long, and pepper them with upper-case letters, numbers and punctuation marks."

Maintaining that the pilots have to be informed at least a month before their scheduled date of operation for an international flight, he said the winter schedule, beginning in October, has to be cleared by aviation regulator Director General of Civil Aviation (DGCA).
The national carrier had been operating 38 out of 45 international flights as part of its contingency plan, which has been extended till August. These flights are being operated by about 120 executive pilots, who are in the management cadre.

Al Qosi, who was born in Sudan around 1960, left in 1996 to join Osama bin Laden in Afghanistan, where he provided services to bin Laden and other al-Qaida members as a driver, bodyguard and cook. In the early 1990s, he trained with jihadists and worked as an accountant for a company affiliated with Osama bin Laden, according to DOD documents released by WikiLeaks.
Al Qosi had been sentenced to a 14-year term for crimes committed between 1996 to 2001, but served two years in exchange for cooperating with prosecutors. The U.S. had agreed to return al Qosi to Sudan upon completing two years of his sentence.
"Although the United States had the legal authority to continue holding al Qosi under the [Authorization for the Use of Military Force], we coordinated with the Government of Sudan on appropriate security measures to mitigate any threat he continues to pose," said Defense Department spokesman Lt. Col. Todd Breasseale in a statement to msnbc.com.
Paul Reichler, a Washington, D.C.-based lawyer who represented al Qosi pro bono for the past seven years, said his client will participate in a re-entry program designed by the Sudanese government for former detainees.
According to a document published by the government in 2010, nine Sudanese nationals had been returned from Guantanamo and been subject to the re-entry program. At that time, none were known to "have engaged in hostilities against the United States, its interests or its allies since their return to Sudan."?
"I?m very glad that he?s a free man," said Reichler, who added that he would have withdrawn his representation of al Qosi if at any time he seemed to be a terrorist or a threat to the U.S. According to al Qosi's court statement, he had no knowledge or or participation in the 1998 Tanzania and Kenya embassy attacks or the September 11 attacks, though he continued to provide logistical support to al-Qaida after these events.
"I believe he is a decent and honorable person whose only desire is to go home to his family, live in peace and tranquility and engage in productive labor in his family business, and he has no desire to be associated with violent movements of any kind," said Reichler.
Al Qosi arrived in the capital city of Khartoum Tuesday evening Eastern time, and according to court documents, will live with his wife, two daughters and other family members upon returning. In a letter to the Military Commission in January 2011, al Qosi's mother and father said that he would manage a family shop in the town of Atira.
One-hundred and sixty-eight detainees remain at Guantanamo. Reichler, who does not represent any other Guantanamo clients, said that many detainees might be interested in negotiating a plea agreement, but that there has been a high degree of skepticism that the U.S. would honor its word.
With al Qosi's release, Reichler said, "I suspect that there will be many who will seek plea agreements."

Ouya Android game console raises $2 million in under 24 hours ...
The Ouya is a $99 video game console designed to connect to your TV. It runs Google Android and features a wireless controller, letting you play Android games on a big screen.
There are some big names attaced to the project including OLPC and Jambox designer Yves Behar and game industry veteran Julie Uhrman, so the project has raised a bit of buzz. Now it?s also raised a lot of money.
Yesterday the folks behind Ouya took to Kickstarter in hopes of raising $950,000 to get the product off the ground. In less than 24 hours, backers had pledged well over $2 million.
As of 11:10 this morning, the project had passed the $2.8 million mark.
Folks aren?t just throwing money at the Ouya. Around 20,000 people have pledged $95 or more to reserve one of the first game consoles once they start shipping in March, 2013. A smaller number has pledged $699 or more to get one of the developer edition models which could ship as soon as December, 2012.
The developers had originally promised to send units to the first 10,000 people that pledged $99 or more ? but that figure has been extended to 80,000? for now.
Since the project has 28 days to go until the fundraising campaign ends, the Ouya folks are looking at ?stretch goals.? Basically, since the original goal was quickly smashed, we could see additional software, hardware, or other features added.
The team is taking suggestions at?ideas@ouya.tv. There?s also a survey to see which games users would most like to see made available in the Ouya game store.
As of now, the Ouya is a box with an NVIDIA Tegra 3 quad-core processor, 1GB of RAM, 8GB of storage, HDMI, WiFi, Bluetooth, and USB 2.0.?It runs Android 4.0.

Fish appear to steer with magnets
Scientists have zeroed in on the likely source of some animals' sense of direction. Rainbow trout seem to be guided by an 'internal compass' of sorts.

When cells scraped from the trout's nasal passages were placed in a rotating magnetic field, a clump of tiny iron-rich crystals inside the cells called magnetite ? the same mineral used in?compass?needles ? spun in synchrony with the field, turning the cells around with them.
The strength of the crystals' magnetic response, and their firm attachment to the surrounding cell membranes, lent strong support for what scientists have long suspected: That these crystals lean back and forth like a sail in response to Earth's weak magnetic field, and that the cells they are embedded in somehow convey their swaying movements to the brain. This is believed to confer trout and other migratory animals with a "magnetic sense" by which to judge direction.
As detailed in a new paper published online July 9 in the journal Proceedings of the National Academy of Sciences, the researchers found that the magnetic cells in the trout's noses swayed in response to a magnetic field 100 times more forcefully than had previously been predicted. "More importantly, we show for the first time that the internal compass needle has a strong connection to the plasma membrane [or outer membrane] of the cell, which is important to realize an immediate sensing process," said lead researcher Michael Winklhofer of the University of Munich in Germany.
The results show that the magnetic cells "clearly meet the physical requirements for a magnetoreceptor" capable of rapidly detecting small changes in Earth's magnetic field, the researchers said.
The strength of Earth's field varies in a predictable way across the planet's surface, allowing migratory animals to use it for position-finding. By learning the strength of a field that exists at a particular destination, the animals can home in on it. That much is pretty well established in the science field; what has remained mysterious is?how?these animals use magnetic-field changes to navigate. [What If Earth's Magnetic Poles Flip?]
Scientists think?Earth's magnetic field?might urge migratory animals in the right direction like a guiding hand pressing on them. "I think it is similar to touch or pressure. The magnetite-based magnetic sense is innervated by the trigeminal nerve, which mediates touch (heat, cold and pain). If the inner compass needle of a cell points in a certain direction in space, and the fish makes a 90-degree turn, the cell will fire and tell the brain: 'I am 90 degrees out of my preferred direction,'" Winklhofer told LiveScience.

Sprint's Galaxy S III update removes local search abilities
Yesterday we heard that the Sprint Galaxy S III was in the midst of getting a small "security" update over-the-air, but with little detail of what exactly was changed. The jury is still out about any security fixes, but one thing that was changed is that you can no longer use the Google search bar to search for local content on the device. This comes on the heels of yet another legal battle between Apple and Samsung, where the unified search ability was enough to get the Galaxy Nexus on a do-not-sell list for about a week.
Luckily, it's pretty easy to fix by using the Google search bar app from a previous version. If you've received the update and miss your universal search, hit the link below for the AC forums thread for details on getting it back.
Samsung, Sprint, we get it. You were forced to make these changes. But calling it a security update and not filling in your users about the full scope of what was being changed on their phones is dirty pool. We expect, and deserve, a little better.

Forget Real Life! New Study Contradicts Existence of ?Facebook Depression?
Despite ominous warnings about "Facebook Depression," an allegedly new mental illness of depression and anxiety caused by overuse of social media, a new level-headed scientific study reveals that social networks have not made 27% of teenagers chronically depressed. Researchers Lauren Jelenchick and Dr. Megan Moreno concluded that there was no association between feeling blue and Facebook use -- even heavy facebook use -- contradicting last year's Academy of Pediatrics report warning parents to monitor their children's social networking activity. "Our study is the first to present scientific evidence on the suggested link between social-media use and risk of depression," said Jelenchick.
